[Confocal] SD: dichroic exchanged for simultaneously imaging CFP/YFP, instead of GFP/RFP

Dear all, The 509 beamsplitter is now installed allowing to simultaneously image CFP/YFP. If you would like to simultaneously image GFP/RFP the beamsplitter needs to be changed to the 580 beamsplitter. This is physically taking out the filter and put the other one in, something that needs to be done by Daniel or me. Keep in mind that before using both cameras you ALWAYS need to calibrate first. This can be done through a userfriendly wizard 'Calibrate Multicamera' (under Video). If not clear, don't hesitate to ask for help. Dear all, The cameras of the spinning disc are exchanged as the rear camera (the mostly used one) gives a very pixeled image. The top camera is now on the place of the rear camera, but settings are for the moment still as top camera. With some adaptations you can use it as normal. Please contact me if you cant to use it, so I can help you with this. [Confocal] SP2: transmission light source brakes rather fast

Dear SP2 users, The transmission light of the SP2 seems to break rather fast (already 2 lamps replaced in the last month, which is exceptional). It can be solved by replacing the lamp. Please come to Daniel or me as we can easily exchange it. If we are not around, you can also ask Kristof (techhelp). A working transmission light gives a visible light at the left side of the microscope (see indicated black box at picture below). You only need this for visualizing your sample through the microscope (NOT for confocal imaging). [Confocal] Leica SP8: sometimes problem with microscope settings

Dear all, The Leica SP8 gives sometimes problems when looking through the microscope to your sample: there is less light and you don't get your sample nicely in focus. It can be solved by clicking (several times) on live view (in the LAS X software), and/or by switching the lens, and/or switching between DIC and POL light (microscope setting). Closing and restarting the PC sometimes helps, but not always, and it is very time consuming. So, I would go for the above options. I assume that there is some kind of filter that gets blocked. Keep this in mind when doing your imaging especially if you are doing treatments, as it could take a bit more time to set up your imaging. I will keep you updated. [Confocal] Leica SP8: please inform me if we still have errors

Dear all, Could you please inform me and register if you still would encounter errors with the LasXsoftware? The upgrade of the software has been done, hoping that most of the issues we had will be solved with this. If not, please inform me. Also register the problem with the error report (LasX software - Help- Create error report), take a printscreen of the error, .... Please save it all on S:\research\images\images_cameras\confocal\Problems_Leica SP8\ (make subfolder with data and name) Hoping that you can enjoy a relaxing imaging time on the SP8!! [Confocal] tomorrow intervention from Leica SP8, for sure whole morning

Dear all, Tomorrow we will have an intervention from Leica SP8. They will start probably around 08:30u and will for sure need the morning. The upgrade of the software will be done, hoping that part of the software errors would be solved with that. (not sure if this will be the case, but this is the first step to do. Looking into these software errors only makes sense with an upgraded software.) Also the repair part for the mirrors for the microscope will hopefully be installed tomorrow (if transport goes according to shedule). I know this is very short notice and this will interfere with your experiments, which I really regret. But I don't see another option. I've delayed this software upgrade already a lot as I wanted to shedule this without interfering the users, but this is a challenging task. Sometimes last-minutes intervention pops up if the Leica people need to be at the IRC site, and they try to combine this with an intervention at our place, which is now the case. Please realize that we are out of guarantee period and we need to pay for each intervention, including transport. A combined visit with IRC is for us also interesting. Lucas, Isabelle, Klaas, Most probably your reservation tomorrow could be taken by the intervention, depending on how it goes. I keep you update.
