[Confocal] SD: dichroic exchanged for simultaneously imaging CFP/YFP, instead of GFP/RFP

Dear all, The 509 beamsplitter is now installed allowing to simultaneously image CFP/YFP. If you would like to simultaneously image GFP/RFP the beamsplitter needs to be changed to the 580 beamsplitter. This is physically taking out the filter and put the other one in, something that needs to be done by Daniel or me. Keep in mind that before using both cameras you ALWAYS need to calibrate first. This can be done through a userfriendly wizard 'Calibrate Multicamera' (under Video). If not clear, don't hesitate to ask for help. [Confocal] Leica SP8: sometimes problem with microscope settings

Dear all, The Leica SP8 gives sometimes problems when looking through the microscope to your sample: there is less light and you don't get your sample nicely in focus. It can be solved by clicking (several times) on live view (in the LAS X software), and/or by switching the lens, and/or switching between DIC and POL light (microscope setting). Closing and restarting the PC sometimes helps, but not always, and it is very time consuming. So, I would go for the above options. I assume that there is some kind of filter that gets blocked. Keep this in mind when doing your imaging especially if you are doing treatments, as it could take a bit more time to set up your imaging. I will keep you updated. Dear confocal users, Could you register overnight sessions by booking the real time you will use it ? (so NOT by reserving till 20:00 and typing ON!) Reason is that the booking is the only calculation we have to convince the management that the machines we have are adequately used. It is therefore in everyone's interest to make sure that these numbers are as close to reality. I do assume nobody will come to shut down the lasers at 2am... If you don't have enough credits, you can ask your groupleader to make some reservations. (every groupleader gets extra credits to cover this).
