[Confocal] SD: dichroic exchanged for simultaneously imaging CFP/YFP, instead of GFP/RFP

Dear all, The 509 beamsplitter is now installed allowing to simultaneously image CFP/YFP. If you would like to simultaneously image GFP/RFP the beamsplitter needs to be changed to the 580 beamsplitter. This is physically taking out the filter and put the other one in, something that needs to be done by Daniel or me. Keep in mind that before using both cameras you ALWAYS need to calibrate first. This can be done through a userfriendly wizard 'Calibrate Multicamera' (under Video). If not clear, don't hesitate to ask for help. [Confocal] SD; transmitted light shutter doesn't work, but imaging is possible with minor adaptations

Dear spinning disc users, The shutter for transmitted light doesn't work. But luckily we are able to have the light coming to our sample, meaning that normal imaging still can be done. But make sure when you switch to confocal mode to have the transmitted light off by using the external box (whereas before you could do this by using the shutter). If you forget this, than all the transmitted light will go on to your detector, which will be dramatic for the camera!! In case of questions, don't hesitate to contact Daniel or me. [Confocal] SD: other joystick, no dual camera, please report problems from coming weeks

Dear spinning disc users, Here are some temporary changes to our spinning disc setup: - other joystick - No dual camera setup possible These adjustments are the result of installation of the loan proscan unit. The purpose is to check if the crashes of the volocity software are due to our proscan unit or not. So it is very important to know how the system will behave in the coming period. So PLEASE report and register ALL PROBLEMS ( print screen, session error log, ....)! And inform me about it. As usual you can save this under \psb.ugent.be\shares\research\images_cameras\spinning disc\Problems Spinning disc I will keep you updated about any change. Hoping that the dual camera setup will soon be available. By mounting your sample on the microscope, this shutter could be accidently pushed up. Dear Olympus users, @ Ignachio, can you confirm as you are the last user today. (I could not find you in the lab) Please follow the instructions in picture below or in attach in order to switch ONLY the 559 and fluorescence lamp off, so LEAVING THE ARGON LASER ON!! Reason is the following: The argon laser took this morning more than one hour to get properly working. THis, together with the non -working 515 laser, makes me thinks that the argon laser is at his end. >From experience, not getting the laser on is often one of the first signs. Once it's on, it's all OK. That's why I adjust the switching off procedure by leaving the laser on! It is a bit risky, with a possible lockdown in prospect, but I don't want to take the risk of not being able to activate the laser on Monday. The reservations are full for next week, and possibly these are experiments that will be finalized next week. Do switch off the 559 and the fluorescence lamp!
