Calendar Name: seminars Scheduled for: Thursday, December 2 2010, 11:00 - 12:30 Event text: Pascal Genschik Institut de Biologie Mol�culaire des Plantes du CNRS IBMP-CNRS - UPR2357 Strasbourg FRANCE Details: "CUL4-DDB1 is required to maintain epigenetic gene repression via its interaction with the Arabidopsis PRC2 complex" ABSTRACT Protein ubiquitylation regulates a broad variety of biological processes in all eukaryotes. Recent work identified a novel class of cullin-RING ligase (CRL) composed of CUL4, DDB1, and one substrate receptor also called DCAF (DDB1 and CUL4-associated factors). DCAFs are WD40 proteins containing WDxR motifs that are required for efficient binding to DDB1. In both human and Arabidopsis, about 90 different DCAFs have been predicted, but for most of them their function or substrates remain still unknown. Among putative Arabidopsis DCAFs we identified MSI1, which belongs to an evolutionary conserved protein family. MSI1-like proteins contribute to different protein complexes, including the epigenetic regulatory Polycomb Repressive Complex2 (PRC2). Here we will present evidence that Arabidopsis MSI1 physically interacts with DDB1 and is part of a multimeric CRL4-type E3 complex. Interestingly the CUL4-DDB1MSI1 complex is required to maintain parental imprinting of MEA, a target gene of the Arabidopsis PRC2 complex. Moreover, we extended these studies to another MSI1-like protein, called MSI4/FVE, which is involved in the regulation of flowering time. Overall, we will discuss the mechanism by which CRL4s are required to maintain Polycomb-mediated gene repression.

Seraing BELGIUM Details: "Use of bacterial poison antidote genes as tools: from DNA engineering to synthetic biology" ABSTRACT Since the �90s, bacterial poison/antidote genes have been used as tools in molecular biology. Today, the ccdA, ccdB gene loci of the F plasmid are present in vectors for protein expression and DNA cloning. These genes are either used for selection of recombinant molecules (Gateway vectors, TOPO vectors, Zero Background vectors) and for the maintenance of expression vectors in E.coli. Nowadays most of the scientists use those genes as tools for their daily research works. Most of the time scientists use those genes ignoring their origin and properties. This lack of information could limit the efficiency of their DNA cloning and protein expression experiments. The aim of this scientific meeting is to present the natural origins antidote poison genes, their properties and open a discussion on their current and future applications.

They have been recently recognised as a novel plant hormone controlling shoot branching. For several years, we have been using garden pea (Pisum sativum) and a series of high branching mutants to study the control of axillary bud outgrowth and how strigolactones, auxin and cytokinin interact to regulate shoot branching. To investigate the presence and the role of strigolactones in a non-vascular plant, we have used the moss Physcomitrella patens, an experimental system well adapted to study the evolution of developmental mechanisms in plants. Using homologous recombination, we have obtained a knock-out mutant in the homologue of the CAROTENOID CLEAVAGE DIOXYGENASE 8 (CCD8) gene, encoding a strigolactone biosynthesis enzyme. The mutant phenotype is rescued by exsudates from wild type colonies, by exogenous supply of synthetic strigolactones or by ectopic expression of seed plant CCD8. Our data demonstrate P. patens produces strigolactones which act as signalling factors controlling both developmental and ecophysiological processes.
