From redox signalling to iron sulfur metabolism and back

Stress-related responses and several developmental processes in plants are accompanied by the formation of reactive oxygen species (ROS) and subsequent translation of this primary oxidation to changes in protein thiol oxidation as a key step in stress signalling pathways. To understand signalling mechanisms, one key feature that needs to be understood is the dynamics of the respective redox changes in time and space. The use of genetically encoded probes for H2O2 and the glutathione redox potential (EGSH) over the past two decades has provided access to the respective physiological parameters in live cells. Genetic dissection of dynamic changes in these parameters including production and transmission systems, allows in vivo testing of hypotheses for protein functions derived from in vitro studies. Glutaredoxins (GRXs) are generally considered key elements for the transmission of redox signals from the local EGSH to protein thiols. Yet, despite a largely conserved overall structure, a second subgroup of GRXs does not function as oxidoreductases but rather acts in iron-sulfur cluster transfer. Beyond addressing the role of GRXs in redox signalling, I will also present our recent work on the mitochondrial iron-sulfur cluster assembly and transfer machinery, which provides new insights into the flux of iron-sulfur clusters and a key role of lipoyl synthase. Alterations in the mitochondrial lipoylation system not only affect mitochondrial metabolism, but may also lead back to cellular redox signalling.
