[Confocal] Leica SP8: sometimes problem with microscope settings

Dear all, The Leica SP8 gives sometimes problems when looking through the microscope to your sample: there is less light and you don't get your sample nicely in focus. It can be solved by clicking (several times) on live view (in the LAS X software), and/or by switching the lens, and/or switching between DIC and POL light (microscope setting). Closing and restarting the PC sometimes helps, but not always, and it is very time consuming. So, I would go for the above options. I assume that there is some kind of filter that gets blocked. Keep this in mind when doing your imaging especially if you are doing treatments, as it could take a bit more time to set up your imaging. I will keep you updated. Dear all, The Leica SP8 can be used again. But without the option to visualize your sample at the microscope with brightfield. Either fluorescence or directly through confocal imaging. However the problem is detected. The sensor to switch the mirrorin the microscope is broken. This part will be ordered and renewed. Estimation is that this would be beginning next week. Gr, Evelien

Dear all, For info: adding extra light (from a normal light source or from mobile phone) on your sample can help you to visualize your cells on the microscope. An normal light source is put next to the microscope for this purpose. Please don't leave it on when you are not there. (to avoid touching the lamp the microscope). Gr, Evelien

Dear Leica SP8 users, We have again the issue with the brightfield light at the microscope that is not working. Leica glued the broken part, but I guess that will not have hold very long. The company will be contacted, and I will keep you up to date as usual. For the moment, you could visualize your samples at the microscope either with fluorescence light or with putting external light above the microscope. (the lamp is for the moment in use by someone working at the olympus confocal, but when needed you can take it from there). Gr, Evelien

We have again the issue with the brightfield light at the microscope that is not working. Leica glued the broken part, but I guess that will not have hold very long. The company will be contacted, and I will keep you up to date as usual.
For the moment, you could visualize your samples at the microscope either with fluorescence light or with putting external light above the microscope. (the lamp is for the moment in use by someone working at the olympus confocal, but when needed you can take it from there).

For info: adding extra light (from a normal light source or from mobile phone) on your sample can help you to visualize your cells on the microscope. An normal light source is put next to the microscope for this purpose. Please don't leave it on when you are not there. (to avoid touching the lamp the microscope).

The Leica SP8 can be used again. But without the option to visualize your sample at the microscope with brightfield. Either fluorescence or directly through confocal imaging. However the problem is detected. The sensor to switch the mirrorin the microscope is broken. This part will be ordered and renewed. Estimation is that this would be beginning next week.

The problems with visualizing your sample through the microscope on the Leica SP8 got worse. We now also have the situation that we have no light at all at the microscope. Switching between live view (in the LAS X software), and/or by switching the lens, and/or switching between DIC and POL light (microscope setting), and the shutters sometimes helps. Restarting the PC doesn't seem to help much, but you can always try.
