Calendar Name: seminars Scheduled for: Friday, October 12 2012, 14:00 - 15:30 Event text: Dr Arp Schnittger Department of Molecular Mechanisms of Phenotypic Plasticity Institut de Biologie Mol�culaire des Plantes du CNRS Universit� de Strasbourg Strasbourg FRANCE Details: �How to make stable decisions in cell proliferation and differentiation� ABSTRACT The decision to replicate its DNA is of crucial importance for every cell and in many organisms decisive to progress through the entire cell cycle. Using pollen as a model system, we have identified a general regulatory cascade controlling entry into S phase in many if not all Arabidopsis cells. This cascade comprises the Arabidopsis homologs of the animal transcription factor E2F, the plant homolog of the animal transcriptional repressor Retinoblastoma (Rb)-related 1 (RBR1), the plant-specific F-box protein, F-BOX-LIKE 17, the plant specific cyclin-dependent kinase (CDK) inhibitors KRPs, as well as CDKA;1, the plant homolog of the yeast and animal Cdc2+/Cdk1 kinases. Biomathematical simulations and subsequent experimental confirmation of computational predictions revealed that this regulatory circuit can give rise to hysteresis highlighting the identified dosage sensitivity of CDK inhibitors in this network. Remarkably, many components of this cell cycle control module appear to function in the control of asymmetric division and cell fate specification as well. I will present data from our team showing that RBR1 not only controls cell proliferation but also cell differentiation genes during root meristem and stomata development. This leads to a CDKA;1-dose dependent model of formative divisions in which low and medium levels of CDKA;1 are sufficient for symmetric divisions but high kinase levels being required for asymmetric divisions. An important aspect of this wiring is again the hysteretic behavior of the system revealing an intimate connection between cell cycle progression and cell fate acquisition.

Plants must have effective means of adapting to changes in their environment. The main features of such defense strategies involve early recognition and perception of the developing stress, and subsequent activation of induced adaptive and defensive responses. Strong evidence has accumulated that ROS play an important role in the signaling under both abiotic and biotic stresses. The air pollutant ozone generates reactive oxygen species (ROS) in the apoplast. Consequently, ozone has been used as a tool to unravel in planta ROS-induced processes and apoplastic ROS sensing. Examples of identification of the molecular identity of some components, and interaction between multiple regulatory cascades involving apoplastic ROS, discovered with this approach are presented.

Calendar Name: seminars Scheduled for: Friday, September 21 2012, 14:00 - 15:30 Event text: Dr Edouard Pesquet Umea Plant Science Centre Department of Plant Physiology Umea University Umea SWEDEN Details: �Mechanisms controlling xylem vessels secondary cell wall patterning and synthesis� ABSTRACT Xylem is essential for transporting water and minerals as well as for mechanical resistance of plants against gravity. These key characteristics of xylem vessels are enabled by three consecutive developmental events which lead to i) the development of specific secondary cell walls which exhibit different type of patterned thickening, ii) the programmed cell death (PCD) to hollow out the cell content and iii) the lignification to reinforce and waterproof its wall for optimal sap conduction. Disturbance of any of these processes greatly impairs xylem vessels function. Using a novel in vitro xylogenic system derived from stable Arabidopsis thaliana cell lines, the mechanisms controlling xylem vessel secondary cell wall patterning and synthesis are studied using a combination of live-cell imaging, pharmacological treatments and genetic studies. Cortical microtubules, through two microtubules interacting proteins MAP70-1 and MAP70-5, control the patterning of xylem vessel secondary cell wall whose deposition is pursued long after cell death.
