'' Transporter function in photoassimilate partitioning and importance for plant performance '' Prof Mechthild Tegeder School of Biological Sciences Washington State University USA Tuesday, May 30, 2023 14:00 Plant growth and development largely depend on the long-distance transport of carbon, nitrogen and sulfur assimilates from photosynthetically-active source leaves to sinks. Sucrose represents the main transport form of carbon, while amino acids are the dominant organic nitrogen and sulfur transport compounds in most plants. Leaf-to-seed allocation of photoassimilates occurs via the phloem and requires loading of the metabolites into the leaf phloem and, at the sink end, their import into the growing embryo. Our research concerns the identification and characterization of key transport processes regulating assimilate partitioning in Arabidopsis and legumes (i.e., pea and soybean) and understanding of their importance for source to sink physiology. Data will be presented demonstrating that cellular transport systems are fundamental in nitrogen, sulfur, and carbon movement from cell to whole plant level and that their function is essential in controlling metabolism and development of source and sink. '' Evolution of plant architecture through changes in auxin movement control '' Dr Yoan Coudert Laboratory of Plant Development and Reproduction ENS Lyon FRANCE Thursday, May 25, 2023 11:00 The successful colonization of land by plants was accompanied by the diversification of their branching architecture. The phytohormone auxin is a major regulator of branch initiation and has a similar inhibitory role in flowering plants and mosses, two major land plant lineages that diverged from their most recent common ancestor several hundred million years ago. PIN-mediated polar auxin transport is crucial for auxin function in flowering plant branching control. Long-range tropic auxin gradients are sustained locally through the regulation of cell-to-cell connectivity at the level of plasmodesmata, although this is comparatively a minor pathway. In the moss Physcomitrium patens, an extant representative of early land plants, PIN proteins have a minor role in leafy shoot branching control and the symplasmic pathway could instead represent the main route for auxin movement in the stem. Using a combination of developmental genetics and computational modeling, we explore the role of symplasmic fields and plasmodesmal gating in auxin movement, and thereby assess their contribution to the evolution of plant morphogenesis. Dubrovsky Department of Plant Molecular Biology Institute of Biotechnology National Autonomous University of Mexico (UNAM) MEXICO Monday, May 22, 2023 11:00 Root system possess a modular organization, in which the root apical meristem (RAM) maintenance and root growth are essential for new root formation. In my lab we study the RAM maintenance and lateral root (LR) formation, and I will discuss our recent discoveries and novel understanding of some processes related to the root system formation. In search of new regulatory pathways of RAM maintenance, we isolated several mutans that are incapable of root growth. This work revealed a new metabolically-dependent regulatory pathway involved in RAM maintenance. Specifically, we established that THREONINE SYNTHASE 1 (TSY1) is critical for stem cell activity independently of the classical SHR-SCR, PLT , and WOX5 regulatory pathways. Using mathematical modeling we were able to reproduce in silico determinate root growth in loss-of-function mutants and predict that not only synthesis but also catabolism of Thr is essential for the RAM maintenance. This work also demonstrated that the RAM proliferation and transition domains can be uncoupled. The RAM function is fundamental for the new root axis and LR root formation, and in the second part of my seminar I will present new insights that show that LR does not start from a preselected group of cells, but in most cases from a single founder cell (FC). We established that after specification of the first FC, a dynamic and non-stereotypic process of FC recruitment from neighboring pericycle cells is taking place. FC recruitment occurs in longitudinal and tangential directions and culminates in the formation of a morphogenetic field of FCs. The recruitment of phloem-pole pericycle to FCs and the role of parent root tissues in the LR formation will also be discussed. I will present our recent data that show how parental root procambium in the vicinity with the FCs participates in the formation of the xylem bridge underlying vascular continuity between all root types in the root system. Although Portugal holds promising food legumes genetic resources, a reduction in local production and consumption in the country, as well as in Europe most traditional markets occurs. Reduced investment in breeding, particularly in nutritional/organoleptic quality, resulted in overall unattractiveness of current varieties not only to farmers/processors but also to consumers and to this national resource underuse. To increase food legumes consumption and cultivation, breeding objectives and market preferences should be better aligned. With the development of high-throughput genetic and phenotypic technologies, it is now possible to address breeding in a more integrated way. At the PlantX Lab, we collect and integrate multiple layers of biological information (metabolomics, phenomics, genomics) through statistical genomics, assuring an improved understanding of this complex biological system, and higher prediction accuracy for target phenotypes, such as yield and quality (through genotype-to-phenotype prediction models). During this presentation, I will revise the work developed on grass pea agronomic, biotic and abiotic stress resistance genetic basis and introduce the more recent nutritional and organoleptic quality studies on the same species. The developed molecular toolbox based on these prediction models will be important for food legumes genotyping, product screening (e.g. quality assessment of legume batches), and marker-assisted selection for routine implementation of quality breeding objectives within the more often focused agronomic improvement. Ubiquitin family proteins are small modifier proteins that are conjugated to proteins and lipids and serve as targeting signals in endosomal and autophagosomal trafficking processes. Thus, for the proper regulation of endosomal and autophagic protein degradation, the spatiotemporal regulation of ubiquitylating and deubiquitylating enzymes as well as ubiquitin-binding proteins is essential. Prof Isono her group is interested in the molecular mechanisms of the regulation and recognition of ubiquitin and ubiquitin-like proteins in intracellular trafficking. They have identified deubiquitylating enzymes (DUBs) that are important for fine-tuning the ubiquitin- and endosomal sorting complex required for transport (ESCRT)-dependent degradation of plasma membrane proteins. Furthermore, they have found proteins that interact with the ubiquitin-like protein ATG8 on autophagosomes and could be key for the recruitment of the ESCRT machinery to non-endosomal membranes. '' P-bodies and post-transcriptional gene regulation in plant reproduction and stress response '' Dr Karel Riha CEITEC Masaryk University Brno CZECH REPUBLIC Thursday, March 16, 2023 11:00 Gene expression is a multilayer process that can be regulated at the level of RNA production, processing, transport, translation and RNA stability. Many of these processes are associated with membraneless organelles, dynamic structures formed by multivalent interactions of proteins and nucleic acids that may exhibit properties of liquid-liquid phase condensates. In his presentation Dr Karel Riha will explore the role of P-bodies, cytoplasmic biocondensates sequestering translationally repressed mRNA and RNA decay proteins, in plant germline differentiation and stress response. He will show how dynamic behavior of P-bodies contributes to regulation of meiosis and discuss a broader function of these structures in other plant physiological processes.
