'' Dr Véronique Jorge UMR BioForA INRAE ONF FRANCE Thursday, December 21, 2023 11:00 Black poplar (Populus nigra L.) is among the tree species having the largest natural distribution in Europe, from West Great Britain to West Asia, and from the foot of the Ural Mountains to the North of Africa. Thus, this species is potentially subjected to a wide variation of climatic conditions. Previous studies on variability of traits in this species showed structure and different distribution patterns depending on the trait, demonstrating potential local adaptation. In the last decade, we have developed molecular tools to study the factors shaping the diversity of the species. Today, these tools allow us to detect patterns of selection along the genome, to identify polymorphism involved in adaptative traits, to model demographic history of the species, and to evaluate how gene flow impact structure of populations. These molecular tools and results constitute the toolbox for breeding and conservation programs of black poplar. It is taken up by plants in the form of inorganic phosphate (Pi). How eukaryotic cells ‘measure’ intracellular Pi levels and respond to changes in Pi availability remains to be uncovered at the mechanistic level. We have previously reported that eukaryotic SPX domains are cellular receptors for Pi-rich inositol pyrophosphate (PP-InsP) nutrient messengers. In Pi sufficient growth conditions, PP-InsP levels are high and enable the binding of the nutrient messenger to its SPX receptor. In plants, the activated receptor can in turn bind a family of phosphate starvation responsive transcription factors rendering them unable to bind DNA. Under phosphate starvation PP-InsP levels drop, the receptor – transcription factor complex dissociates and the released transcription factor can activate Pi-starvation responsive gene expression. I will present our current mechanistic understanding of this nutrient signaling pathway as well as a quantitative biochemical, structural and physiological characterization of PP-InsP biosynthesis and catabolism in the model plants Arabidopsis and Marchantia. We discovered that some sRNAs from eukaryotic pathogens, such as Botrytis cinerea, can be transported into host plant cells and suppress host immunity genes for successful infection (Weiberg et al., Science 2013). We further demonstrated that plants can also send sRNAs into pathogens, mainly using extracellular vesicles, mostly exosomes, to silence fungal virulence genes as part of its immune responses (Cai et al., Science 2018). We identified a group of RNA binding proteins that contribute to the selective sRNA loading into extracellular vesicles (He et al., Nature Plants, 2021). Furthermore, we recently demonstrate that B. cinerea also utilizes extracellular vesicles to secrete sRNAs, which are then enter plant cells through clathrin-mediated endocytosis (He et al., Nature Communications, 2023). Thus, extracellular vesicles play an important role in cross-kingdom RNA trafficking between plants and fungal pathogens. In addition, we discovered that environmental RNAi is present in many fungal pathogens, which can take up RNAs from the environment efficiently (Wang et al,, Nature Plants, 2016; Qiao et al., Plant Biotechnology Journal 2021). Applying small RNAs or double-stranded RNAs that target fungal virulence-related genes on plants can effectively inhibit fungal diseases. Innovative pathogen gene-targeting RNA-based antifungal solutions using nanotechnologies represent a new generation of fungicides that are durable and eco-friendly (Qiao et al., Plant Biotechnology Journal, 2023; Niño-Sánchez et al., Journal of Integrative Plant Biology 2023). This ability is particularly striking during root development where plants have to forage in highly heterogeneous environments. I will describe how plant hormones enable roots to sense and/or respond to environmental signals. Examples include discovering how plants sense availability of moisture in soil by linking intercellular water fluxes with movement of hormones like auxin and ABA, triggering changes in root branching designed to maximise capture of soil resources (Orosa et al, 2018, Science; Mehra et al, 2022, Science). Plant roots also employ volatile signals like ethylene to sense changes in soil physical properties like compaction stress using a novel gas diffusion based mechanism (Pandey et al, 2021, Science). I will conclude by describing how mechanistic insights about hormone-regulated root plasticity is helping design stress resilient crops. Through the recruitment of damaged and unwanted cellular components into de novo formed double membrane vesicles, known as autophagosomes, and their subsequent degradation in lytic compartments, selective autophagy facilitates cellular reprogramming. The recruitment of specific cargo is mediated by cargo receptors that bridge the cargo with the growing autophagosome. Despite recent advances that underscore the critical role of selective autophagy in stress tolerance, our understanding of the full range of cargo receptors involved in these processes remains incomplete. In this presentation, I will describe our recent findings, where we leveraged evolutionary diversity to discover new cargo receptors in plants. Our comparative studies in plants have led us to identify a cross-kingdom conserved selective autophagy process that maintains endoplasmic reticulum homeostasis in humans. Through this work, we hope to establish autophagy as a template for addressing a fundamental question in evolutionary cell biology: how do cellular innovations arise in different branches of life? '' ATP production in mitochondria through respiration and its use for protein homeostasis in plants '' Prof Harvey Millar ARC Centre of Excellence in Plant Energy Biology The University of Western Australia Perth AUSTRALIA Monday, October 30, 2023 15:00 Cellular respiration is a foundational biochemical process linking the oxidative decarboxylation of organic acids to the generation of ATP for use in biosynthesis and cellular maintenance. I will outline recent work we have been conducting to understand the rate of this process. This includes determining the transport routes for primary organic acid substrates that plant mitochondria use to generate ATP and also new insights into the efficiency of ATP generation inside plant mitochondria through the electron transport chain. I will then outline a systematic means using proteomics technology to measure the investment of this ATP through protein synthesis to build the steady-state (but constantly turning over) metabolic machinery of the plant cell. Using progressive labelling approaches with stable isotope we have studied changes in energy use patterns during high light and during periods of darkness in Arabidopsis through varying rates of protein synthesis and protein degradation of specific enzymes and biochemical pathways. I will outline how we are using these insights in research focused on crop plants in the field, the emerging requirements for vertical farming and plant-based protein production, right through to new opportunities for plants as critical systems in provisioning future space exploration. Collectively these approaches aim to return us from the current re-occupation in molecular plant science with amounts of components to consider more fully the rates of processes that drive growth and plant productivity. '' Mechanisms controlling polar localization and boron-dependent degradation of boric acid/borate transport proteins '' Prof Junpei Takano Laboratory of Plant Nutrition Graduate School of Agriculture Osaka Metropolitan University JAPAN Thursday, September 28, 2023 11:00 Boron (B) deficiency and toxicity are agricultural problems impeding crop production worldwide. Due to relatively high solubility of B as boric acid (B[OH]3), B deficiency often occurs in high rain fall areas, while B toxicity occurs in arid and semi-arid areas. Boron is essential for plant growth and survival due to its function in cross-liking pectin at the rhamnogalacturonan II (RG-II). Under low-B conditions, a boric acid channel NIP5;1 and a borate [B(OH)4–] exporter BOR1 play key roles in B uptake and translocation in Arabidopsis thaliana. In root cells including epidermal and endodermal cells, NIP5;1 and BOR1 are localized to the plasma membrane in polar manners toward the soil- and stele-side, to support directional transport of B toward the root stele. The polar localization of NIP5;1 and BOR1 is mediated not by a static retention mechanism, but by constitutive endocytosis and exocytosis dependent on phosphorylation in N/C-terminal regions. It is likely that different modes of transcytosis are operating for the polar localization toward the soil- and stele-side cell domains. When the B concentration in soil fluctuates, plants have to control activities of these transport proteins. Upon sufficient-B supply, BOR1 undergoes polyubiquitination and is transported from the plasma membrane to the vacuole for degradation, to avoid overaccumulation of B. Our forward genetic screen identified that amino acid residues located in vicinity of the substrate-binding pocket of BOR1 are essential for the vacuolar sorting. BOR1 variants that lack B-transport activity showed a significant reduction of polyubiquitination and subsequent vacuolar sorting. Our study on BOR1 and other studies on yeast amino acid transporters suggest that ubiquitination of these transporters relies on its conformational transition during the transport cycle. In Arabidopsis, characteristic PCD events are crucial for the correct development of the lateral root cap, the most external tissue of the root. The lateral root cap translates the environmental stimuli into internal cellular response, thus leading to the regulation of the growth and architecture of the entire organ. In order to ensure tissue proper development and functions, the uppermost cells of this tissue undergoes a characteristic PCD event. Recent evidences hinted on a correlation between the activity of the hormone auxin and PCD events, including those occurring in the lateral root cap. Exploiting a live imaging approach combined with cell type specific gene editing, we show that auxin transport dynamics govern the timing of the uppermost lateral root cap cells PCD occurrence. We thus suggest the existence of a specific molecular circuit that confers positional cues to lateral root cap cells to coordinate cellular turnover and whole organ development.
