“Building eco-responsive strategies to manage plant parasitic nematodes” Prof. Baldwin Torto International Centre of Insect Physiology and Ecology (icipe) Nairobi, Kenya Thursday, May 23, 2024 11:00 Semiochemicals mediate chemical communication and interactions between and within species. Examples include sex and aggregation pheromones that have been investigated and identified largely for above-ground insect pests. For below-ground pests including plant parasitic nematodes, chemical cues have been found to play crucial roles in host and non-host plant discrimination, essential for survival of various species. Understanding the nature and identity of these chemical signals offers opportunities for developing effective eco-responsive solutions for sustainable agriculture, especially for smallholder farmer productivity. In this presentation, I will highlight advances made in our group to identify semiochemicals that mediate specific behaviors in plant parasitic nematodes. Towards developing a resilient management strategy for plant parasitic pests, I will draw examples from our work based on laboratory and field experiments to i) elucidate the mechanisms of host finding in the root knot nematode Meloidogyne incognita, and use of non-host companion crops to suppress this phenotype in tomato, and ii) use of a biodegradable lignocellulose matrix to disrupt chemical signalling mediating host finding in the invasive plant parasitic nematode, the potato cyst nematode Globodera rhostochensis. '' Single-cell driven enhancer modelling and design '' Prof Stein Aerts VIB Center for AI & Computational Biology KU Leuven BELGIUM Tuesday, April 30, 2024 11:00 am The combination of scRNA-seq and scATAC-seq allows building gene-regulatory atlases of any tissue and species. I will present several new computational strategies that exploit single-cell multi-omics data: (1) to model genomic enhancers using topic modelling and convolutional neural networks; and (2) to derive “enhancer-GRNs” (eGRN) with key transcription factors, genomic enhancers, and predicted target genes per cell type. I will discuss the results of several case studies where we applied these strategies, including the Drosophila brain, human melanoma, the mouse liver, and the evolution of cell types in the vertebrate telencephalon. Finally, I will discuss how enhancer models based on deep learning can be exploited to design synthetic enhancers for Drosophila and human cell types. '' Control of plant phosphate homeostasis by inositol pyrophosphate nutrient messengers '' Prof Michael Hothorn Department of Plant Sciences Faculty of Science Science III University of Geneva SWITZERLAND Thursday, March 7, 2024 11:00 Phosphorus forms an essential structural component of membranes and nucleic acids, represents an important signaling molecule and the energy currency of the cell. It is taken up by plants in the form of inorganic phosphate (Pi). How eukaryotic cells ‘measure’ intracellular Pi levels and respond to changes in Pi availability remains to be uncovered at the mechanistic level. We have previously reported that eukaryotic SPX domains are cellular receptors for Pi-rich inositol pyrophosphate (PP-InsP) nutrient messengers. In Pi sufficient growth conditions, PP-InsP levels are high and enable the binding of the nutrient messenger to its SPX receptor. In plants, the activated receptor can in turn bind a family of phosphate starvation responsive transcription factors rendering them unable to bind DNA. Under phosphate starvation PP-InsP levels drop, the receptor – transcription factor complex dissociates and the released transcription factor can activate Pi-starvation responsive gene expression. I will present our current mechanistic understanding of this nutrient signaling pathway as well as a quantitative biochemical, structural and physiological characterization of PP-InsP biosynthesis and catabolism in the model plants Arabidopsis and Marchantia. However, the mechanisms that underlie this important redox-based modification remain poorly understood in higher plants. We have, therefore, analyzed how protein persulfidation acts as a specific and reversible signaling mechanism during the abscisic acid (ABA) response in Arabidopsis thaliana. We show that ABA stimulates the persulfidation of L-CYSTEINE DESULFHYDRASE 1 (DES1), an important endogenous H2S enzyme, at Cys44 and Cys205 in a redox-dependent manner. Moreover, sustainable H2S accumulation drives persulfidation of the NADPH oxidase RESPIRATORY BURST OXIDASE HOMOLOG PROTEIN D (RBOHD) at Cys825 and Cys890, enhancing its ability to produce reactive oxygen species. Physiologically, S-persulfidation-induced RBOHD activity is relevant to ABA-induced stomatal closure. Together, these processes form a negative feedback loop that fine-tunes guard cell redox homeostasis and ABA signaling. These findings not only expand our current knowledge of H2S function in the context of guard cell ABA signaling, but also demonstrate the presence of a rapid signal integration mechanism involving specific and reversible redox-based post-translational modifications that occur in response to changing environmental conditions.
