'' MAP Kinase Signaling in Cell Polarity - a Lesson from the Plant Embryo '' Prof Martin Bayer Center for Plant Molecular Biology (ZMBP) Developmental Genetics University of Tübingen GERMANY Thursday, January 26, 2023 11:00 Breaking symmetry by asymmetric cell divisions is essential for establishing different cell identities in multi-cellular development. Cell-cell signaling by receptor kinase/MAP kinase signaling pathways seems to be a reoccurring mechanism in polarizing plant cells and establishing different cell identities in the daughter cell of asymmetric cell divisions. We are using the early plant embryo as system to study initial events in cell polarization by the ERECTA-YODA signaling pathway – a prototype receptor kinase/MAP kinase signaling pathway. Embryogenesis resembles a linear system of developmental progression with successive asymmetric cell divisions that produce the primary tissue types of the plant seedling and establish the stem cell niches of the shoot and root. By comparing the function of the ERECTA-YODA pathway in the embryo with its role in planar patterning of the leaf epidermis, we identified principal functions of a core pathway and context-specific modifications. We present new data on the impact of polar YODA activation on early embryonic patterning and shed light on the mechanism and evolution of distinct modes of YDA activation in the zygote on a molecular and structural level. We furthermore discuss possible benefits of different modes of YDA activation and their distinct parent-of-origin effects. Ubiquitin family proteins are small modifier proteins that are conjugated to proteins and lipids and serve as targeting signals in endosomal and autophagosomal trafficking processes. Thus, for the proper regulation of endosomal and autophagic protein degradation, the spatiotemporal regulation of ubiquitylating and deubiquitylating enzymes as well as ubiquitin-binding proteins is essential. Prof Isono her group is interested in the molecular mechanisms of the regulation and recognition of ubiquitin and ubiquitin-like proteins in intracellular trafficking. They have identified deubiquitylating enzymes (DUBs) that are important for fine-tuning the ubiquitin- and endosomal sorting complex required for transport (ESCRT) - dependent degradation of plasma membrane proteins. Furthermore, they have found proteins that interact with the ubiquitin-like protein ATG8 on autophagosomes and could be key for the recruitment of the ESCRT machinery to non-endosomal membranes. '' Auxin signaling: more than we could ever imagine '' Prof Jiri Friml Institute of Science and Technology Austria (ISTA) Klosterneuburg AUSTRIA Friday, October 14, 2022 14:00 The plant hormone auxin is a versatile intercellular signal influencing virtually all aspects of plant life. It has a unique ability to be directionally transported within tissues forming local auxin maxima or gradients that are central to many developmental processes mediated by auxin. One of the key roles of auxin is adaptation of plant growth to gravity, where shoots bend up and roots down. This paradox is based on opposite responses of these organs to the phytohormone auxin, which promotes cell expansion in shoots, while inhibiting it in roots via an unclear signalling pathway and yet unknown downstream cellular mechanism. The well-established canonical auxin signalling involving the TIR1/AFB auxin receptors, Aux/IAA repressors and ARF transcription factors acts in nucleus and mediates gene transcription. However, auxin also triggers cellular responses within seconds or minutes, too fast to rely on transcription. Part of the rapid responses is mediated by the non-transcriptional branch of the TIR1/AFB signalling, but others involve a yet completely unknown mechanism. Here Prof Jiri Friml will present new and surprising insights into the mechanism of auxin signalling including an ultrafast auxin-triggered protein phosphorylation response and previously unsuspected aspects of TIR1/AFB auxin perception and downstream signalling. '' MicroRNAs in plants: biogenesis and roles in plant growth and development '' Prof Javier Palatnik Institute of Molecular and Cellular Biology of Rosario (IBR ) ARGENTINA Monday, September 26, 2022 11:00 MicroRNAs are essential endogenous regulators of gene expression in plants. They are generated from foldback precursors by the microprocessor, whose main component is DICER-LIKE1. The mature miRNAs of ~21 nt length are incorporated into ARGONAUTE complexes, which they then guide to target mRNAs that are subsequently cleaved or translationally arrested. In contrast to their stereotypical animal counterparts, plant microRNA precursors are very heterogeneous in size and shape. We have discovered that plant microRNAs can be generated by several different modes, explaining the variability of precursors that can generate these small RNAs. Many evolutionarily conserved plant microRNAs regulate transcription factors involved in plant development. MicroRNA miR396 regulates GROWTH-REGUALTING FACTORS (GRFs), which are plant-specific transcription factors. The microRNA miR396-GRF module has been implicated in the promotion of organ growth and as a regulator of crop yield. We have been studying the integration of the miR396 regulatory network in the developmental program of the plant. Interestingly, while miR396 regulates cell proliferation in leaves, in the root apical meristem it aids to control and maintain the stem cell population. A general view of the microRNA biogenesis pathway, as well as the roles of the miR396 regulatory network will be presented. ''Deciphering the role of brassinosteroids in plant adaptation to climate change'' Dr Ana Caño Centre for Research in Agricultural Genomics (CRAG) Barcelona SPAIN Wednesday, March 30, 2022 11:00 Despite the massive amount of information gathered around the functions and mechanisms of Brassinosteroids (BRs) in plants, an important limitation persists in our knowledge of this signaling pathway: almost all we know comes from observations on the BRI1 receptor pathway, that is essential for growth and development, and for which mutants are highly pleiotropic and typically dwarf. Since the discovery of BRI1-like receptors (BRL1/3) (1) , we still do not really grasp what are their fundamental functions in plants. Twenty years of research (by ours and other labs) have resumed the analysis of BRLs as redundant BRI1 receptors with a marginal vascular expression and lack of apparent mutant phenotypes. Strikingly, our work carried in the context of an ERC CoG “ICRICA” takes a novel perspective to explore the function of BRLs in Arabidopsis, to understand the inner working of this pathway. In light of our recent findings showing that overexpression of BRL3-receptors confers drought resistance, we have now deciphered the BRL3 pathway in Arabidopsis, including novel components in the pathway essential to plant adaption to climate change. Our new data not only change the paradigm for our present understanding of BR signaling in plants, but also open new and exciting avenues for engineering climate resilient crops. The latest results of our research will be presented at the VIB seminar.
