[Confocal] olympus confocal: IMPORTANT!! switching off adapted procedure!!!

Dear Olympus users, @ Ignachio, can you confirm as you are the last user today. (I could not find you in the lab) Please follow the instructions in picture below or in attach in order to switch ONLY the 559 and fluorescence lamp off, so LEAVING THE ARGON LASER ON!! Reason is the following: The argon laser took this morning more than one hour to get properly working. THis, together with the non -working 515 laser, makes me thinks that the argon laser is at his end.
From experience, not getting the laser on is often one of the first signs. Once it's on, it's all OK. That's why I adjust the switching off procedure by leaving the laser on! It is a bit risky, with a possible lockdown in prospect, but I don't want to take the risk of not being able to activate the laser on Monday. The reservations are full for next week, and possibly these are experiments that will be finalized next week.
Do switch off the 559 and the fluorescence lamp! See cadaped switching off procedure below: Gr, Evelien Gr, Evelien Evelien Mylle - Labmanager/ microscopy technician Advanced Live Cell Imaging VIB-UGent Center for Plant Systems Biology Ghent University Technologiepark-Zwijnaarde 71 - 9052 Ghent - Belgium Tel. +32(0)9 331 39 74 [ http://www.psb.ugent.be/ | www.psb.ugent.be ] Genome editing, cutting-edge technology for a sustainable agriculture

Hi Jelle, thank you for letting us know! I checked now, the argon laser is in principle on, but the 488nm emission is very, very weak (barely visible by eye at 100%) and I have not yet been able to switch to 515nm. I have not yet started the experiment, but seeing the emission, I am not even sure I will, if I get any result I will let you know. I think it is pretty safe to say the argon laser is in a terminal coma, if not already gone :( Matous Matouš Glanc - Postdoctoral fellow Vascular Development VIB-UGent Center for Plant Systems Biology Ghent University Technologiepark-Zwijnaarde 71 - 9052 Ghent - Belgium Tel. +32(0)9 331 39 45 [ http://www.psb.ugent.be/ | www.psb.ugent.be ] Genome editing, cutting-edge technology for a sustainable agriculture

Dear olympus confocal users, The argon laser is giving up its last forces: at the moment there is only a very minimal output, hardly impossible to image something. The malfunctioning 515 laser, the difficult start up of the argon laser were the first signs of decay. The laser has been ordered for a while, but for now there is no news when it will be delivered and placed. So now is the ideal time for imaging with the 405 and 559 laser. I will keep you further informed. Greetings, Evelien

Dear all, The olympus confocal is running at full power again, with a new argon laser. The previous one had 6147 hours, which can be considered a 'beautiful age' and understandable that he needs eternal rest. Dear confocal users, I find it very disappointing to see that very important instructions are not followed by some users. One of the criteria for accessing a confocal is to turn the system on and off correctly, and thus be up to date when changes occur. Determining that this is missing is something that I find very serious. There has been a very explicit request not to turn off the argon laser. This has been communicated via email and there is also a note on the laser itself. It has happened twice since then that the laser has been turned off anyway, which meant that the next user had to wait a half to full day before the laser was turned on. So I don't know what the cause of the not following is: not being informed (not registered in the confocal mailing list?), ignoring notes on the device,...? Suggestions to have everyone follow these instructions are always welcome! Best regards, Evelien
